Accurate oil level reading

Hi all, am just about to embark on a long journey on my Innova, can anyone tell me how to get an accurate oil level reading? Do you just dip the dipstick in or do you screw it in before unscrewing it out again? I might be a dipstick asking such a question so forgive my ignorance, better to be safe than sorry
