From my experience with vintage stuff where the reg number has been lost.
You have to get the bike mot'd using the frame number as the registration then book an appointment with your local DVLA vehicle examiner.
You can then apply for an age related reg number.
Unless of course the bike is nicked and the frame number shows up on the dvla computer, in which case it will be all for nothing and will be seized and crushed.
